Enzymatic cascade systems for D-amino acid synthesis: progress and perspectives

نویسندگان

چکیده

D-amino acids, different from the ubiquitous L-amino are recognized as “unnatural” amino acids. The applications of acids have drawn increasing interest researchers in recent years, and widely used various industries, including for food products, pharmaceuticals, agricultural chemicals. Inspired by prevalent applications, many synthetic methods been developed, which mainly divided into chemical biosynthetic methods. Chemical synthesis has a variety disadvantages such multiple reaction steps, low yields, rates, difficulties product extraction. Thus, utilizing enzymes attracting attention because they more energy-saving environmentally friendly compared to traditional synthesis. Among all enzymatic methods, multi-enzymatic cascade catalytic significant advantages, lower costs, no need intermediate separation, higher efficiency, is ascribed spatial proximity biocatalysts. In this review, advances multi-enzyme systems well chemo-enzymatic approaches synthesize discussed.

Non-targeted Identification of D-Amino Acid-Containing Peptides Through Enzymatic Screening, Chiral Amino Acid Analysis, and LC-MS.

D-Amino acid-containing peptides (DAACPs) in animals are a class of bioactive molecules formed via the posttranslational modification of peptides consisting of all-L-amino acid residues. Amino acid residue isomerization greatly impacts the function of the resulting DAACP. However, because isomerization does not change the peptide's mass, this modification is difficult to detect by most mass spe...

Enzymatic synthesis of desoxyribonucleic acid.

Double stranded DNA serves as a template primer for Escherichia coli DNA polymerase when the DNA contains a single strand break (a nick) with a 3’-hydroxyl terminus. Initiation of replication entails covalent extension of the 3’-hydroxyl terminus and a concurrent 5’ -+ 3’ nuclease action by the enzyme. The primer strand is hydrolyzed at the 5‘ side of the nick while the synthetic activity catal...

Enzymatic Synthesis of Ribonucleic Acid

The discovery of enzymes which catalyze the deoxyribonucleic acid-dependent synthesis of ribopolynucleotides (l-6) stimulated an immediate inquiry into the role played by DNA in thisunique reaction. The speculation that DNA might serve as a template for the polymerization of ribonucleotides into a complementary ribonucleic acid molecule was confirmed by studies from a number of different groups...

Enzymatic synthesis of deoxyribonucleic acid.

Determination of deoxyribonucleotide sequence in a deoxyribonucleic acid molecule is important from both the chemical and genetic points of view. It is also essential for answering the question of whether DNA synthesized in vitro by polymerase (1, 2) is a faithful copy of the nucleotidel sequence of the primer DNA. Although enzymatically synthesized DNA has the same over-all nucleotide composit...

Structural, thermal and enzymatic analysis of naturally occurring and D-amino acid substituted peptides

Designing of new peptide materials for biomedical and protein engineering applications are important. In the present work an attempt has been made to study the effect of D-Leu in collagen like tetra peptide on the structure and stability of peptide against enzymes and results are compared with its chiral counterpart L-form. Effect of replacement of L-Leu in Leu-Gly-Pro-Ala tetra peptide with D-...
